چکیده
Background Why the adaptive immune system turns against citrullinated antigens in rheumatoid arthritis (RA) and whether anti-citrullinated protein antibodies (ACPAs) contribute to pathogenesis are questions that have triggered intense research but still not fully answered. Neutrophils may be crucial this context, as sources of also targets ACPAs. Objectives To better understand how ACPAs neutrophils RA pathogenesis, we studied reactivity a broad spectrum patient-derived ACPA clones activated or resting neutrophils, compared neutrophil binding using CCP2 captured polyclonal from different patients. Methods Human were isolated peripheral blood healthy volunteers, individuals at risk for [1] patients with established RA. The cells Ca 2+ ionophore, phorbol 12-myristate 13-acetate (PMA), nigericin, zymosan IL-8. monoclonal preparations was flow cytometry confocal microscopy. Neutrophil extracellular trap formation (NETosis) analyzed IncuCyteZOOM TM . role arginine deiminase (PAD)2 PAD4 obtained PAD-deficient mice inhibitors. Osteoclast cultures human monocytes presence M-CSF RANK-L. Results targeted NET-like structures all tested conditions did bind surface intact influence NETosis. We observed prominent clonal diversity capacity neutrophil-derived antigens. PAD2 dispensable majority required binding. When patients, high patient-to-patient variability targeting Similar inter-individual another cellular effect ACPAs: stimulation osteoclast differentiation. Conclusion can important under lead activation, NETosis extrusion intracellular material. A substantial among both suggest RA-related symptoms variability. HLA-DRB1 Genotypes and the Risk of Developing Anti Citrullinated Protein Antibody (ACPA) Positive Rheumatoid Arthritis
OBJECTIVE To provide a table indicating the risk for developing anti citrullinated protein antibody (ACPA) positive rheumatoid arthritis (RA) according to one's HLA-DRB1 genotype. METHODS We HLA-DRB1 genotyped 857 patients with ACPA positive RA and 2178 controls from South Eastern and Eastern France and calculated Odds Ratios (OR) for developing RA for 106 of 132 possible genotypes accounting...
متن کاملThe periodontium of periodontitis patients contains citrullinated proteins which may play a role in ACPA (anti-citrullinated protein antibody) formation.
AIM To determine the presence and location (stroma versus epithelium) of citrullinated proteins in periodontitis tissue as compared to non-periodontitis tissue and synovial tissue of RA patients. MATERIALS & METHODS Periodontitis, healthy periodontal and RA-affected synovial tissue samples were collected in addition to buccal swabs. These samples were stained for the presence of citrullinated...
متن کاملContribution of Peptide Backbone to Anti-Citrullinated Peptide Antibody Reactivity.
Rheumatoid arthritis (RA) is one of the most common autoimmune diseases, affecting approximately 1-2% of the world population. One of the characteristic features of RA is the presence of autoantibodies. Especially the highly specific anti-citrullinated peptide antibodies (ACPAs), which have been found in up to 70% of RA patients' sera, have received much attention. Several citrullinated protein...
متن کاملThe fine specificity of IgM anti-citrullinated protein antibodies (ACPA) is different from that of IgG ACPA
INTRODUCTION The antigen recognition pattern of immunoglobulin M (IgM) could, when directed against protein antigens, provide an indication of the antigenic moieties triggering new B cells. The half-life of IgM is short and memory B cells against T-cell-dependent protein antigens typically produce IgG and not IgM antibodies. In this study, we analyzed whether a difference exists between the fin...
متن کاملThe role of anti-citrullinated protein antibodies (ACPA) in the pathogenesis of rheumatoid arthritis
The most specific autoimmunity known for rheumatoid arthritis (RA) is reflected by generation of anti-citrullinated protein antibodies (ACPA). Presence of ACPA in established RA is associated with disease severity, while generation of ACPA at early developmental phases of RA can have a strong predictive value for progressing to the full-blown disease. Hence, development of ACPA may be of crucia...
